Sickle cell disease is a disease of genetic origins, which translates into high cell and tissue oxidative stress in both acute and chronic forms. Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidine N-oxyl) is a chemical used as an antioxidant that mimics superoxide dismutase in scavenging free radicals. This product is used in pharmacology and neuroscience research, but there are no published studies on the use of tempol in treatment of neurodegenerative diseases. We evaluated the effects of tempol in vitro and in vivo on oxidative stress-related parameters, such as reactive oxygen species (ROS), nitric oxide (NO) levels, and MDA levels, and on apoptosis-related parameters, such as mitochondrial membrane potential (ΔΨm), Bax and Bcl2 expression, cytochrome c, cleaved caspase-3, and caspase-3 levels. Our results show that tempol treatment decreases ROS and NO levels, and MDA levels. In addition, tempol also increases ΔΨm, Bcl2 expression, and inhibits the expression of Bax and cleaved caspase-3 in a PC12 cell line exposed to glutamate and in a rat model with quinolinic acid-induced damage. Thus, tempol may have therapeutic potential for the treatment of neurodegenerative diseases by providing a neuroprotective effect.[Abdominal pseudocyst after gastrectomy. Apropos of a case].
The authors report a case of pseudocyst following gastrectomy for cancer of the stomach. In the absence of a gastric pouch after gastrectomy and total gastrectomy, the latter being associated with esophageal stricture or esophageal atresia (Fessler operation), the presence of a pseudocyst must be considered in the differential diagnosis of abdominal abscess. After total gastrectomy, the use of a nonabsorbable clip to prevent pancreatic leakage is not sufficient to isolate a pseudocyst from the pancreatic juice and should be reconsidered.
