The primary sex determining loci, the H-Y antigens, SRY and the second testis-determining gene, ZFY2, have been shown to be tightly linked at one end of the sex chromosome pair in Mus musculus. Southern hybridization analysis of four mice lines containing known homologous substitutions in the sex chromosome region demonstrates that the SRY-containing H-Y chromosomes in the B6D2F1 strain appear to be identical to the chromosome bearing SRY in the other mouse strains examined. Microsatellite genetic markers have defined a minimum set of recombination breakpoints separating the linked H-Y and the SRY loci in B6D2F1. We have examined the association of these primary sex-determining loci with certain components of prolificacy (number of offspring produced), including sperm production, spermatogenesis, embryo production (number of blastocysts produced), and overall successful embryo production. We observed no significant differences in sperm production, in two-cell and eight-cell embryo production, or in any of the aforementioned factors between mice containing the linked H-Y and SRY loci compared with mice containing a B6D2F1-specific form of SRY.
